Overview
ACE-031 is not a small peptide but an engineered fusion protein designed to act as a decoy for the activin receptor type IIB. By capturing myostatin and related ligands before they reach the true receptor, it removes a natural brake on muscle growth. It was investigated in clinical trials, including in Duchenne muscular dystrophy, before development was discontinued over safety signals. It is not an approved medicine.
How it’s thought to work
- Acts as a soluble decoy receptor that binds myostatin and related TGF-beta ligands.
- Prevents these ligands from activating the true activin type IIB receptor.
- Removing myostatin's inhibitory signal is thought to promote muscle growth.
- Also binds other ligands such as activin, which contributed to off-target effects.
Researched uses

Reconstitution
Supplied in clinical trials as a lyophilized protein reconstituted per study protocol; weight-based dosing means simple fixed-unit syringe math does not directly apply.
ACE-031 dosage calculator
The calculator below is pre-filled with ACE-031’s commonly cited vial size and dose. Adjust the vial amount, bacteriostatic water, or dose to match your own vial — it will show the exact insulin-syringe units to draw, the concentration, and how many doses the vial holds.
Add 1 mL of bacteriostatic water to your 1 mg vial. That gives you about 1 doses of 1 mg. For each dose, draw 100 units into a 100-unit insulin syringe.

Cautions
- Not an approved drug; clinical development was halted over safety concerns.
- Trials reported bleeding-related events such as nosebleeds and gum bleeding.
- Off-target activin binding produced effects beyond muscle.
- Prohibited in competitive sport as a myostatin-pathway agent.
